Facilitating Research and Development in Bioprocesses
Time to experiment, clinic, or market is becoming increasingly important for developers using bioprocessing. Shorter timelines, faster development programs, new molecules, and increased regulatory expectations are all challenges that researchers are facing nowadays. Process understanding is the foundation of today’s process development. Having insights into how the different Critical Process Parameters (CPPs) interplay with each other using modern technologies and solutions is key to improving development efficiency. Selecting the correct technologies and the instrumentation for research and development to characterize and monitor the processes is critical to future scale-up success.
METTLER TOLEDO process analytics, spectroscopy, and laboratory technologies and industrial weighing solutions perfectly fit your bioprocessing research and development.
Optimization and Research Steps
The steps from development in small, often transparent, containers to a relevant production volume into large, stainless-steel vessels present the user with the task of creating the same (or at least similar) conditions for the cells as factories. Culture conditions (growth conditions and media components) can be developed and optimized in several small-scale bioreactors. Design of Experiment (DOE) performed in parallel runs with different set permutations allow rapid findings of the optimal fermentation and expression yield conditions. Once parameters have been optimized, the process can be scaled up.
Media Optimization
With more than 50 components in culture media, screening and studying all the combinations to find the ideal formulation is labor-intensive, costly, and time-consuming. The choice of components and parameters—such as temperature, mixing, and pH—can significantly affect media quality and the maintenance of a consistent and performant cell culture.
In-line and offline accurate measurements are performed throughout media optimization. Therefore, process analytical technology (PAT) and design of experiment (DoE) are often necessary to develop a robust process and an ideal media formulation.
Upstream Processing
Developing a robust control strategy relies on a reliable real-time culture monitoring solution. According to the PAT initiative, process analyzers should provide multivariate process-related information using nondestructive, noninvasive measurements.
While in-line monitoring devices for CPPs such as pH, dissolved oxygen (DO or pO2), dissolved carbon dioxide (dCO2 or pCO2) and temperature can be considered the foundation of real-time bioprocess measurements and are well-established, monitoring other variables such as cell density, substrates, and products requires analyzers with particular characteristics.
Also, the use of smart weigh modules or high-precision scales under tanks enables an additional facet of condition monitoring. Active alerts to potential risks help you ensure process success.
Downstream Processing
A successful downstream processing (DSP) process pipeline depends on the quality of the therapeutic design and the upstream processing as well as a cascade of well-designed and operated separation, purification, and formulation strategies.
With the increasing pressure to boost productivity and reduce the cost of goods, scientists are developing processes to enable faster regulation and commercialization of the biomolecules.
They are adopting new technology such as single-use technologies to reduce finished costs and simplify the process, inline process analytical technologies to streamline and integrate the analytical data (mainly pH, flow, pressure and conductivity), weight-based process automation, and process modeling to deepen the process understanding and control and achieve greater process insight on continuous processing methods.
